چکیده

Adoptive cell transfer of tumor-specific T cells provides an effective strategy for cancer immunotherapy. An article in this issue provides a novel approach to refine this technology by identifying tumor-reactive T cells based on frequency and PD-1 expression. Trends in Adoptive Tumor Infiltrating Lymphocytes (TILs) Therapy

Adoptive T cell therapy based on tumor infiltrating lymphocytes (TILs) is the most effective treatment for malignant melanoma. The TILs therapy has demonstrated high overall response rates, resulting in tumor regression and prolonged survival in comparison to IL-2 and ipilimumab treatments. Expansion of TILs under exvivo conditions using patients’ own tumor tissue is a prerequisite for effectiv...

CD137 accurately identifies and enriches for naturally occurring tumor-reactive T cells in tumor.

PURPOSE Upregulation of CD137 (4-1BB) on recently activated CD8(+) T cells has been used to identify rare viral or tumor antigen-specific T cells from peripheral blood. Here, we evaluated the immunobiology of CD137 in human cancer and the utility of a CD137-positive separation methodology for the identification and enrichment of fresh tumor-reactive tumor-infiltrating lymphocytes (TIL) or tumor...

PD-1 identifies the patient-specific CD8⁺ tumor-reactive repertoire infiltrating human tumors.

Adoptive transfer of tumor-infiltrating lymphocytes (TILs) can mediate regression of metastatic melanoma; however, TILs are a heterogeneous population, and there are no effective markers to specifically identify and select the repertoire of tumor-reactive and mutation-specific CD8⁺ lymphocytes. The lack of biomarkers limits the ability to study these cells and develop strategies to enhance clin...

Expansion of tumor-infiltrating lymphocytes and their potential for application as adoptive cell transfer therapy in human breast cancer

Adoptive cell transfer (ACT) of ex vivo expanded tumor-infiltrating lymphocytes (TILs) has been successful in treating a considerable proportion of patients with metastatic melanoma. In addition, some patients with several other solid tumors were recently reported to have benefited clinically from such ACT. However, it remains unclear whether ACT using TILs is broadly applicable in breast cance...
